It's only 1 line (the bold line in the middle). And that hanger is only big enough to handle 1 helo. Of course the destroyer can carry 2 helicopters but 1 will be sitting out in the open. I'm sure the reasons why ships have hangers is so they can work/maintain the chopper after each flight; put on gears and other stuff. Having it sit on the landing pad will be inconvenient to do all those stuff.

Oh i was not referring to that line
I was referring to the other 2 lines, with a hole which i think is used to assist in directing/moving the helicopter into the hangar. One of them has an equipment as you can see in the picture you showed.

They can, but depending on how big those are. Akizuki's are indeed capable of storing two SH-60K or J in the aircraft hangar though I never seen any pictures of that as that would be considered emergency and there aren't too many pictures of them taken during big manouvers. Asahi class on the other hand also can take two in the hangar but one must be operated by hand as there's only one rail so in case of Asahi class we can assume that a second one would be stored there in reserve as it wouldn't be fully operational in case of real crisis.
